Abstract
One of the biggest bottlenecks in transplantation medicine and biomedical engineering is the long-term preservation of biological tissues and even organs. Even though hypothermic storage has become a regular practice in clinic, its restriction on the availability of organs is severely restricted by the limited viability duration. This difficulty could be beaten by cryopreservation, but the efficacy is limited by two closely interconnected variables: the diffusion of cryoprotective agents (CPAs) through tissues and the cytotoxicity of these agents. Dimethyl sulfoxide (DMSO) is among the most frequently used permeating CPAs, since it aids in avoiding the formation of intracellular ice, but its toxicity also rises with concentration, which is one of the main control factors when developing and optimizing protocols.
The relationship between CPA transport and DMSO-induced toxicity in murine heart and kidney tissues is examined in this thesis by conducting experiments at various scales
