Abstract:

Alzheimer's Disease (AD) stands as one of the most impactful neurodegenerative diseases worldwide. This study aims to delve into one of its primary hypotheses: the Tau hypothesis. According to this theory, AD primarily stems from the hyperphosphorylation of the protein MAP Tau, causing it to detach from Microtubules, thereby disrupting them and leading to neuronal death. In further detail, ongoing research is expected to lead to the future classification of proteins involved in Tau phosphorylation by analyzing geometrical descriptors calculated on the meshes. Utilizing computational tools, molecular dynamic simulations were conducted on nine systems each representing a Microtubule protofilament stabilized by the Tau protein's Microtubule-binding domain, exhibiting various combinations of phosphorylated serine sites. Another crucial aspect is elucidated through the interfaces between Tau and Microtubules. This research verifies the theory that the greater the phosphorylation of MAP Tau is, the more pronounced its detachment from Microtubules is. (as evidenced by the diminishing value of the area between Microtubules and Tau)
